SAMPLE PREPARATIONS

全血/血清/血漿 Whole blood/ Serum/Plasma	Directly add sample in anticoagulant vacutainer to 50 ul PCR reaction mixture.	1) Add lysis solution. 2) Incubate the tubes until completed lysis is achieved. 3) Spin down briefly. 4) Directly use 1 ul supernatant of crude lysate per 50 ul PCR reaction. * Some samples requires previous preparations such as centrifugation & collection of the supernatant before 1). * Lysis Buffer I is for SDS lysis. * Lysis Buffer II is for Alkaline lysis.
培養細胞 Cultured Cells	Suspend cells from a plate and/or wash them with PBS, and directly add 1ul of cell suspension per 50 ul PCR reaction.
口内細胞 Buccal Cells	Immerse the brush into 10 ml PBS, and directly use 1 ul solution per 50 ul PCR reaction.
唾液 Saliva	Add 0.25 - 10 ul of saliva to 50 ul PCR reaction mixture.

パラフィン埋包組織切片 Paraffinized Tissue slide/TMA	After deparaffinization, scratch up the tissue of interest with syringe needle. Immerse the edge of the needle covered with tissue powders to 50 ul PCR reaction mixture and gently shake.

毛根 Hair root	Use Lysis Method
バクテリア/ウィルス Bacteria(Gram-)/ Virus from feces	Suspend sample in PBS and crush well, centrifuge at 3,000 g for 1min and collect the supernatant, and directly use 1 - 10 ul solution per 50 ul PCR reaction.
喀痰 Sputum	After liquefied, centrifuge and resuspend pellet in 50 ul of 1x Elution Buffer. Directly use 1 ul solution per 50 ul PCR reaction.
気管支洗浄 Bronchial lavage 液体/尿 fluid/Urine	Centrifuge and resuspend the sample in 50 ul of 1x Elution Buffer. Directly use 1 ul solution per 50 ul PCR reaction.
バクテリア Bacteria	For some gram negative bacteria such as E.coli, directly add one colony to 50ul PCR reaction with sterilized toothpick.
植物/真菌/酵母 Plant/Fungus/Yeast	Use Lysis Method
乾燥血液/紙に付着した口内細胞 Dried blood/Buccal cells on the papers	Punch a disk out of the sample area on the FTA Card, and then place 1-2 disk spot in a PCR tube and add PCR master mix directly to the disk and amplify.	N/A
